Should the battery be fully charged on 16 SR while PT is being installed to ensure cell balance?

If I had to take a guess, I would say that both the bike's battery pack and the PT should be fully charged and at as close as possible to the same voltage.

The main pack and the power tank each have a contactor and BMS; the contactors won't close unless the voltages match (within 1V say? I thought I had that worked out but can't see where I documented it).

I'd estimate that the safest way to join the two is to connect the PT harness, then plug in the onboard charger to bring the lower pack voltage to match the higher pack voltage at which point the open contactor should close. But I have only seen PT tests performed by others and my understanding may be fuzzy.
